    Target Protein

Principal Names: ACHE; YT; acetylcholinesterase (YT blood group)
    Official Symbol: ACHE
    Accession Number(s): NP_000656.1
    Human GeneID(s): 43
Gene Ontology Terms: DNA replication; acetylcholine binding; acetylcholine catabolism in synaptic cleft; acetylcholinesterase activity; amyloid precursor protein metabolism; basal lamina; beta-amyloid binding; cell adhesion; cell proliferation; cholinesterase activity; extracellular region; hydrolase activity; membrane; muscle development; neurotransmitter catabolism; positive regulation of protein secretion; protein homodimerization activity; response to wounding; serine esterase activity; synapse; synaptogenesis
Important Comments: This antibody is expected to recognise isoform NP_000656 only (the ubiquitously expressed, hydrophillic form).

    Immunogen

Peptide with sequence QFDHYSKQDRCSDL, from the C Terminus of the protein sequence according to NP_000656.1.

    Purification + Storage

Purified from goat serum by ammonium sulphate precipitation followed by antigen affinity chromatography using the immunizing peptide.
Supplied at 0.5 mg/ml in Tris saline, 0.02% sodium azide, pH7.3 with 0.5% bovine serum albumin.
Aliquot and store at -20°C. Minimize freezing and thawing.

    Applications Tested

Peptide ELISA: antibody detection limit dilution 1:64,000. Western Blot: Approx 70kDa band observed in Human Brain (Cerebellum and Hippocampus) lysates (calculated MW of 67.8kDa according to NP_000656.1). Recommended concentration: 0.3-1µg/ml.
An additional band of 150kDa was consistently observed, however this band was not blocked by the immunizing peptide and it is therefore a non-specific signal. We call for caution when used for other assays than Western blot.

    Species Reactivity

    Tested: Human
Expected from sequence similarity: Human, Mouse, Rat, Cat, Rabbit

    Background Reference

Cottingham MG, Voskuil JL, Vaux DJ.
The intact human acetylcholinesterase C-terminal oligomerization domain is alpha-helical in situ and in isolation, but a shorter fragment forms beta-sheet-rich amyloid fibrils and protofibrillar oligomers.
